Multicomponent reactions (MCRs) are the processes involving at least three polyfunctional reagents and leading to the rapid assembly of complex molecular architectures, can be recognized as one of the most effective methods in organic synthesis. The Biginelli reaction used for the synthesis of 3,4-dihydropyrimidin-2-ones by a three-component condensation of urea, aromatic aldehydes and β-ketoesters is one of the most studied and well-known MCRs. Various modifications of this reaction provide a simple and variable route to the synthesis of many nitrogen-containing heterocycles, which are a core structural element of many drugs.
