A seminar “Modifications of the Biginelli reaction for the synthesis of various nitrogen-containing heterocycles” will be held at RUDN University.

Multicomponent reactions (MCRs) are the processes involving at least three polyfunctional reagents and leading to the rapid assembly of complex molecular architectures, can be recognized as one of the most effective methods in organic synthesis. The Biginelli reaction used for the synthesis of 3,4-dihydropyrimidin-2-ones by a three-component condensation of urea, aromatic aldehydes and β-ketoesters is one of the most studied and well-known MCRs. Various modifications of this reaction provide a simple and variable route to the synthesis of many nitrogen-containing heterocycles, which are a core structural element of many drugs.

This presentation is dedicated to the modification of the Biginelli reaction for the synthesis of trisubstituted imidazoles from readily available acyclic N, N-bis-nucleophiles, 2-oxoaldehydes and 1,3-dicarbonyl compounds. Most of the synthesized products were successfully used in the subsequent photocyclization reaction to form structurally interesting polycyclic molecules. The replacement of N, N-bis-nucleophiles to ketenaminals allowed to carry out a similar process in a chemoselective manner to obtain a diverse library of fused polyheterocycles including pyrrolo[2,1-b]thiazoles, pyrrolo[1,2-a]imidazoles and imidazo[1,2-a] quinolines.
